Nonuniform distribution of fibronectin during avian limb development☆
[摘要] Usingindirectimmunofluorescencewehaveexaminedthedistributionofthecellsurfaceandextracellularmatrixglycoproteinfibronectinattheepithelial-mesenchymalinterfaceandinthemesenchymeofdevelopingchickandduckwingbuds.Atallstagesexamined,inbothspecies,stainingforfibronectinisgreatlyenhancedinthebasementmembranesubjacenttotheapicalectodermalridge(AER),asiteofinductivetissueinteraction,relativetotheepithelialbasementmembranesinthenoninductivedorsalandventrallimbepithelial-mesenchymalinterfaces.Instage23,25,and28chicklimbbuds,stainingforfibronectinisuniformintheleastmaturedistalmesenchyme,retainedbetweenmoreproximalcellsundergoingprecartilagecondensationandlostinthoseregionsundergoingmyogenesis,andpersistentinallbutthemostmaturecartilagepresentatthelateststageexamined.TheseresultsareconsistentwitharoleforfibronectininAER-inducedlimboutgrowth,andwithatransientrolefortheglycoproteinintheformationoftheskeletalpatternofthelimb.
