Survival of isolated, fully grown mouse ovarian oocytes is strictly dependent on external Ca2+☆
[摘要] ThepresenceofCa2+isessentialforsurvivalincultureoffullygrownoocytesisolatedfrommouseovariesbutnotforsurvivalofsmall,meioticallyincompetentoocytes,metaphaseIIoocytes,andearlyembryos.Ninetypercentoffullygrownovarianoocytesdiewithin2hrwhenculturedincalcium-freemedium(CFM).CFMdeathdoesnotoccurifothercations(1mMLa3+or10mMSr2+,butnot12mMMg2+nor1mMD-600)replaceCa2+inthemedium.SensitivitytoCFMisprogressivelyacquiredbytheoocyteduringthegrowthphase,inparallelwiththeacquisitionofmeioticcompetence,andislostafter2hrofcultureinthepresenceofatleast0.5mMCa2+.ThelossofsensitivitytoCFMduringinvitrocultureisnotrelatedtotheconcomitantspontaneousresumptionofmeiosis,sincetheoocytebecomesresistanttoCFMevenifgerminalvesiclebreakdownispreventedbytheadditionofdibutyrylcAMPtotheculturemedium.Somehypothesesareputforwardtoexplainthepeculiarandtransienthighcalciumrequirementsoffullygrownoocytes.
