Neuronal survival in intact ciliary ganglia in vivo and in vitro: Ciliary neuronotrophic factor as a target surrogate☆
[摘要] Ciliaryneuronotrophicfactor(CNTF)requirementsforneuronalsurvivalintheintactciliaryganglion(CG)havebeeninvestigatedinorganculture.ExogenousCNTFwasnotessentialforneuronalsurvivaluntilembryonicDay8.Three-dayculturesfrom5-daygangliaweresimilarwithorwithoutCNTF,showingnumerousneuronsandextensiveneuriticdevelopment.In3-dayculturesfrom8-day-oldganglia,however,noneuronssurvivedwithoutCNTF,andthegangliacontainedonlynonneuronalcellsandcelldebris.SimilargangliaculturedwithCNTFcontainedmanyneurons,surroundedbynonneuronalcells,andabundantneuriticprocesses.MorphologicmaturationoftheneuronswaslessadvancedinCNTF-supportedgangliathanintheirinvivocounterparts.
