The control of embryonic pigment cell proliferation in culture by cyclic AMP☆
[摘要] Isolatedpigmentcellsinlong-termculturesderivedfrom14-daymouseepidermalectodermareresponsivetocAMPintheculturemedium.By48hrafterstimulation62%ofthecellsareproliferatingasmeasuredbytheuptakeof[3H]thymidine,comparedto7%incontrolcultures.Bothmelanoblastsandmelanocytesrespond,andwithinashortperiodoftimetheculturescontainlargenumbersofproliferatingpigmentedandunpigmentedcells.Melanocytesstillwithintheepidermalsheetsofshort-termculturesdonotrespondtoaddedcAMP,andtheirnormalrateofproliferationistotallyinhibited.Melanoblastsin14-dayembryonicepidermisthathasbeencompletelydissociatedrespondtocAMPbyahigherplantingefficiencyandincreasedrateofproliferation.TheseresultssuggestthatthecAMPsystemmayplayanimportantroleinthecontrolofmelanoblastandmelanocytenumbersduringdevelopmentwhentheeventsofcellmigrationandtheestablishmentofdensityrelationshipswithintheepidermisareoccurring.
