The effect of experimentally induced triploidy on the lethal expression of the t12 mutation in mouse embryos☆
[摘要] Diploidembryoswhicharehomozygousforthet12mutationdieatthemorulastage.Inthecurrentstudies,ovafromheterozygous(+t12)femaleswerefertilizedinvitrowithspermatozoafrom+t12males.ThefertilizedovawereimmediatelyplacedintomediacontainingcytochalasinBtopreventsecondpolarbodyformation,producing+/+/+,+/+/t12,+/t12/t12,andt12/t12/t12embryos.Thesubsequentdevelopmentofthesetriploidembryoswascomparedwiththatofdiploid+/+,+t12,andt12t12embryosdevelopingfromovawhichwerealsofertilizedinvitrowithspermatozoafrom+t12malesbutwhichwerenottreatedwithcytochalasinBimmediatelyfollowinggametecoincubation.Thedatashowthatthosetriploidembryoswhichpossessawild-typealleleandtwomutantallelesarephenotypicallywildtypewhilethosepossessingthreemutantallelesarenotphenotypicallydistinguishablefromtheirdiploid(t12t12)counterparts.Liket12t12embryos,t12/t12/t12embryosdieatthemorulastage,priortoblastocoeliccavityformation.
