Phosphorylation of the contact site A glycoprotein (gp80) of Dictyostelium discoideum☆
[摘要] DevelopmentallyregulatedcohesionofDictyosteliumdiscoideumcanbeblockedbytheFabfragmentofantiserumpreparedagainstaglycoproteinofabout80,000daltons,gp80,purifiedfromthemembranesofdevelopingamoebae.Immunoprecipitationofgp80withthisserum,from32P-labeledcellextractsofaggregatingD.discoideumamoebaeshowedittobeaphosphoprotein.Serinephosphatewasfoundinthemolecule.Allmultipleisoelectricformsofgp80werephosphorylated.Synthesisofthephosphorylatedformofgp80wasfoundtobelimitedtotheperiodofaggregationandcoincidedwiththeperiodofincorporationof[35S]methionineintogp80.Phosphorylationcouldberapidlyinhibitedbycycloheximidesuggestingthatphosphorylationoccursonlyonnewlymadegp80.Nounphosphorylatedgp80couldbedetectedincellextracts.
