Messenger RNA encoding basic chromosomal proteins of mouse testis☆
[摘要] Considerableknowledgehasaccumulatedconcerninghistonesandotherbasicchromosomalproteinswhichappearinmammalianmalegermcellsduringspermatogenesis.Thebestcharacterizedoftheseproteinsaretestisprotein(TP)andprotamine.Botharesmallandhighlybasic,TPbeingpresentinearlyspermatidswhiletwomolecularspeciesofmouseprotaminemaybedetectedinspermatidsinthelaterstagesofdifferentiationandinsperm.TheaminoacidsequenceofTPfromratandofasinglespeciesofprotaminefrombullhavebeenreported.Incontrasttothisdetailedinformationrelatedtoproteinstructure,relativelylittleisknownofthemechanismsinvolvedinsynthesisoftheproteinsorofthecontrolswhichregulatetheirsynthesisduringspermatogenesis.Inthestudyreportedhere,messengerRNA(mRNA)frommousetestiswasexaminedforitsabilitytoactasatemplateforinvitrosynthesisofTPandprotamine.ThreepolyacrylamidegelelectrophoresissystemsweredevelopedtodistinguishTPandthetwospeciesofmouseprotamineamonginvitrotranslationproductssynthesizedbyacell-freewheatgermextract.A6SRNAencodingTPwasidentifiedbytranslationofsucrosegradientfractions.RNAencodingTPwasalsofoundamongthetestisRNAswhichwereboundtooligo(dT)-celluloseduringaffinitychromatography.ThesephysicalpropertiesandapparentabundanceofthisTPmRNAsuggestthatitmaybeausefulprobeforfurtherstudiesofRNAmetabolismandgeneregulation.Innocasewasaninvitrotranslationproductcomigratingwiththetwoprotaminespeciesobserved,suggestingthepossibilitythatprotaminemaybesynthesizedasaprecursorandlaterprocessedtothespecieswhicharefoundinsperm.
