Membrane potential changes associated with differentiation of enterocytes in the rat intestinal villi in culture
[摘要] Fragmentedvilliofthesmallintestineisolatedfromnewbornratsweremaintainedincultureforperiodsofupto4weeks.Inculture,floatingglobularvilliandadherentvilliweredistinguished.Intracellularrecordingsweremadefrombothtypesofvilli.Themembranepotentialwashighest(about−70mV)inthecellsatthetipofthevillusandwaslowest(about−18mV)inthecellsnearthecryptregion,showingacontinuousgradationaccordingtothecelllocation.ThemembranepotentialinculturedmatureenterocyteswasduetothecontributionoftheelectrogenicNa+pumpaswellastotheCa2+-activatedK+conductance.Boththecomponentsweretemperatureandmetabolicenergydependent.Basedonthesedata,itissuggestedthattheCa2+transportmechanismandtheelectrogenicNa+pumpareinvolvedintheprocessofcelldifferentiationormaturationoftheintestinalepithelia.
