Disease lesion mimics in maize: I. Effect of genetic background, temperature, developmental age, and wounding on necrotic spot formation with Les1☆
[摘要] Les1,adominantgeneofmaize(ZeamaysL.),resultsintheproductionofnecroticleafspots.Expressionofthistraitistemperaturesensitive,andthenonpermissivetemperatureforexpressionisdeterminedbythegeneticbackgroundinwhichthegeneisplaced.Exposuretononpermissiveconditionsfor24–48hrwillinducelesionproductioninthemostsensitivegenotype.Lesionsformfirstattheleaftip,theoldestpartoftheleaf,andprogressbasipetallythroughfullyexpandedtissue.Leavesexcisedfromplantsgrownateitherpermissiveornonpermissivetemperaturesandplacedinwater,gibberellicacid,orabscisicacidsolutionsformnonewlesionsateither20or30°C,andtheleavessenescerapidly.However,whenleavesexcisedfromnormalandLes1plantsareplacedinkinetin,senescenceisdelayedandnumerouslesionsdevelopat20°ConLes1plants.Ourresultssuggestthatthereisadevelopmentaltimewindowduringwhichmaizeleafcellscanbeinducedtoformlesions:cellsmustbefullyelongatedbutnotyetsenescent.Thishypothesisisstrengthenedbytheobservationthatpinprickwoundingofleavesinduceslesionsonlyinabandoftissueapproximately2daysyoungerthantheareaoftheleafcurrentlyproducinglesions.VariousmodelsfortheactionofLes1incausingdiscretelesionformationarediscussed.
