The roles of phenotype and position in guiding the fate of 16-cell mouse blastomeres☆
[摘要] Newlyformed16-cellblastomeresweretypedaslargerorsmaller,labelledwiththeshort-termlineagemarkerFITC,andaggregatedinvariousspatialarrayswith15otherage-matchedunlabelled16-cellblastomeres.Theaggregateswereculturedfor8or24hrandthefluorescentlylabelledprogenyidentified.Inallbut6of185cases,theprogenydevelopedasaphysicallycoherentpatch.Labelledlargercellsplacedontheoutsideoftheaggregategeneratedmainlytrophectoderm;whenplacedontheinsideorrandomlytheyalwaysgeneratedatleastonetrophectodermaloffspringandinsomecasesalsocontributedcellstotheinnercellmass(ICM).LabelledsmallercellsplacedontheinsideoftheaggregategeneratedmainlyICM;whenplacedontheoutsideorrandomlytheygeneratedcellsintheICMalone,introphectodermalone,orinbothtissues.Fromtheseresultsweconcludethatphenotypeisofmajorimportanceindeterminingthefateoflargercellswhereaspositionstronglyinfluencesthefateofsmallercells.
