Temperature-sensitive carrot variants impaired in somatic embryogenesis☆
[摘要] Culturedcellsofmanyplantspeciesarecapableofregeneratingbacktoplants.Incarrots,plantregenerationtakesplaceviaembryogenesis.Becausevastnumbersofsomaticembryosareproducedfromhaploidcarrotcultures,itispossibletodesignaselectionprocedureforrecoveringtemperature-sensitivevariantsimpairedinearlyembryogenesis(ts-emb−).Thets-emb−linesfallintothreeclasses.Thefirstclassisblockedbeforeanygrowthinembryogenicmediumcantakeplace;thesecondclassisblockedinorganizedgrowth:cellscangrowascallus,butcannotformembryos;thethirdisblockedatthefirststageofembryodevelopment,theglobularstage.Theseclassesareusefulindelineatingthedevelopmentalstepsduringearlysomaticembryogenesis.Thets-emb−phenotypeofmostvariantlinesisstableincultureandismaintainedthroughplantregeneration.Hypothesesaremadeconcerningtheoriginofthets-emb−phenotype.Inadditiontothets-emb−lines,otherclassesofvariantshavebeenrecovered,i.e.,ts-growth−,habituatedlines,andlineswithalteredresponsestogrowthregulators.
