The mechanism of cell elongation during lens fiber cell differentiation☆
[摘要] Lensfiberformationischaracterizedbyextensivecellelongation.Earlierstudieshaveshownthatlenscellelongationinvitrocanoccurintheabsenceofmicrotubulesandisassociatedwithaproportionalincreaseincellvolume.Wehavepreviouslysuggestedthatlensfibercellelongationisdirectlycausedbyanincreaseincellvolume.Inthisreport,lensesfrom3-and6-day-oldchickenembryoswerethree-dimensionallyreconstructedfromserialsectionstoprovideameasureofcellvolumeandlengthduringvariousstagesofprimaryandsecondarylensfiberformation.Inbothcases,cellvolumewashighlycorrelatedwithcelllengthduringlenscellelongation.Inaddition,duringprimarylensfiberformation,largeintercellularspacesbetweenlensvesiclecellsdisappearedasthesecellsbegantoelongatetoformlensfibers.Lossofintercellularspaceswouldbeexpectedifincreasingcellvolumewereresponsibleforcellelongation.Finally,resultsofexperimentsinwhichthelenscapsulewascutwithafinetungstenneedlesuggestedthatthecapsulewaselasticandnormallyundertension.Thesefindingswereusedtoformulateamodelwhichaccountsforthemajoreventsinlensmorphogenesisbasedon(1)theregulationofcellvolume,(2)thejunctionspresentbetweenlenscells,and(3)theconstraintprovidedbytheelasticityofthelenscapsule.
