Cellular migration through the cardiac jelly matrix: A stereoanalysis by high-voltage electron microscopy☆
[摘要] Formationandmigrationofcushiontissueinthedevelopingchickheartwasanalyzedbyscanningandhigh-voltageelectronmicroscopicstereoanalysis.Twomethodsoffixationwhichenhancethepreservationofwater-solublecomponentsoftheextracellularmatrix(cardiacjelly)wereemployed:1%tannicacidin3%glutaraldehyde(TAG)and1%cetylpyridiniumchloride(CPC)in3%glutaraldehyde.Ourresultsindicatedthatthepreservationofthecell:matrixinteractionexhibitedbyendocardialcellsandmigratingcushiontissueisdependentuponthemethodoffixation.InTAG-fixedembryos,filopodialextensionsfromtheendocardiumaswellasfilopodiaofpioneeringcellsaremostoftenassociatedwithmicrofibrillarcomponentsofthematrix,whereasinCPC-fixedmaterialthesesamecellularextensionsarefoundinassociationwithpleomorphicanastomosingstrandsrichinhyaluronate.Followingtheseinitialcell:matrixinteractionsbyboththeendocardiumandpioneeringcells,trailingcellsinvadetheextracellularmatricalregionandclearlyencounterinbothtypesoffixationadifferentmicroenvironmentinwhichtoengageincell:matricalassociations.Theseobservationssupportthehypothesisthatfilopodialprobingbyendocardialcellsandpioneeringcellsresultsinmacromolecularreorderingsofthematrixandthussuggestanadditionalfunctionforfilopodiabeyondtranslocationofthecells.
