Channel open time of acetylcholine receptors on Xenopus muscle cells in dissociated cell culture☆
[摘要] Thekineticsofacetylcholine(ACh)receptorchannelsonculturedmyotomalmusclecellsfromXenopusembryoswerestudiedbyanalyzingfocallyrecordedmembranecurrents.Themeanopentimeforreceptorchannelsonembryonicmusclecellsgrownindissociatedcellculturesshowedatime-dependentdecreasesimilartothatseeninvivo.ThechangesinpowerdensityspectraareconsistentwiththehypothesisthatthedecreaseresultsfromtheappearanceofaclassofAChreceptorwithashortmeanchannelopentime(0.7msec)andadecreaseintheproportionofreceptorswithalongmeanchannelopentime(3msec).TheadditionofdissociatedneuraltubecellstomusclecellculturesresultedinanunexpectedincreaseinmeanchannelopentimeforAChreceptorsinbothsynapticandnonsynapticregions.ThesestudiesdemonstratethatAChreceptorfunctionmaybealteredinculturedmusclecells.
