Developmental patterns of cytoplasmic transcript prevalence in sea urchin embryos☆
[摘要] Severalhundredtwo-cellstage,gastrulastage,andpluteusstageseaurchinembryocDNAcloneswerescreenedwith[32P]cDNAtranscribedfromadevelopmentalseriesofcytoplasmicandpolysomalpoly(A)RNAs.TranscriptprevalencewasestimatedbyreferencetothereactionofaseriesofstandardclonescomplementarytoRNAsofknownabundance.Wedescribethedominantpatternofprevalencechangeduringdevelopmentforabundantsequences,i.e.,thosepresentintherange5×104–106molecules/embryo.Thesetranscriptsaremostoftenaboutequallyabundantineggandpluteusstageembryos,andareseveralfoldlessprevalentatgastrulastage.Lessthan10%ofthesequencesdisplayingthispatternundergo>10-foldchangesinprevalenceduringdevelopment.Asearchforlaterembryosequencesnotrepresenteddetectablyineggorcleavagestageembryopoly(A)RNAyieldedanumberofexamples.ThesewereconfirmedbytheRNAgelblotmethod.Sequencesregulatedinthismannerallbelongedtolowerabundanceclasses,andarepresentatabout40copies/cellorlessatthegastrulastage.
