Transcription of Xenopus borealis rRNA genes in nuclei of Xenopus laevis oocytes☆
[摘要] RecombinantplasmidsthatcontainedasinglerRNAgeneandspacerfromXenopusborealiswereinjectedintothenucleiofXenopuslaevisoocytes.Afterspreadingthecontentsofinjectednucleiforelectronmicroscopy,wefoundtranscriptionunitsindicativeoftheaccuratetranscriptionoftheborealisgene.BoththenumberofRNApolymerasemoleculespertranscriptionunitandthetotalnumberofactivegenespernucleusweresimilartothosefoundforinjectedlaevisgenes.WealsoinjectedlaevisoocyteswithaplasmidthatcontainedbothashortenedlaevisrRNAgeneandafull-lengthborealisrRNAgene.Visualizationoftranscriptiononthisplasmidshowedthatusuallyeitherashortoralongtranscriptionunitwaspresentonanyonemolecule.Themorphologiesoftheshortandlongtranscriptionunitswereconsistentwiththeirrepresentingtranscriptionofthelaevisandborealisgenes,respectively.Bothtypesoftranscriptionunitwerepresentinroughlyequalnumbers.SincethelaevisoocytetranscribedinjectedborealisrRNAgenesasefficientlyasittranscribedinjectedlaevisrRNAgenes,itstandsinmarkedcontrasttothesituationinembryosofinterspecifichybridsbetweenlaevisandborealiswheretranscriptionoccursonlyfromthelaevisrRNAgenes.PossibleexplanationsforthedifferenceintranscriptionalbehavioroftheborealisrRNAgeneinhybridembryosandlaevisoocytesarediscussed.
