Monoclonal antibodies recognizing cell surface antigens in Drosophila melanogaster
[摘要] MonoclonalantibodieshavebeenmadeagainstcellsurfaceantigensfromDrosophilamelanogaster,asprobesfor“differentiationantigens.â€Theimmunogensusedwere0–16hrembryoniccellsandmassisolatedimaginaldiscs.Thetissuedistributionsoftheantigensrecognizedbythreeantiembryonicantibodiesandtwoantiimaginaldiscantibodieshavebeendefinedbyindirectimmunofluorescent(IIF)assaysondifferentiatedembryoniccellculturesandondissectedthirdinstarlarvalorgans.Theseantigensfallintotwomajorcategoriesbeingeitherubiquitousortissue-limitedindistribution.Inindirectradioimmunoassaysagainst12Drosophilacelllinestheantigensshowingubiquitoustissuedistributionsweredetectableonallcelllineswhereasthetissue-limitedantigenswereabsentfromsomecelllines.Suchascreenagainstcelllinesthereforeprovidesastraightforwardmeansofidentifyingantibodiesagainstnonubiquitousantigens.Oneantibodyrecognizingatissue-limitedantigenwasdetectedasmuscle-specificbyIIFassaysondifferentiatedembryoniccellcultures.Thesecondtissue-limitedantigenwasfoundtolabelbasementmembranes,byIIFassaysagainstthirdinstarlarvalorgans.Thetemporaldistributionoftheantigensduringembryogenesis(3–21hr)hasbeenstudiedbyindirectradioimmunoassay.Inthisrespecttheantigensfallintothreeclasses:(1)abundantthroughoutembryogenesis(aubiquitousantigen),(2)presentthroughoutembryogenesisbutincreasingmarkedlyinabundancebetween12and15hr(twoubiquitousantigensandthemuscle-specificantigen),and(3)absentearlyindevelopmentbutappearingatabout12hrpostfertilization(thetissue-limited,basementmembraneantigen).
