A 28K-dalton cell surface autoantigen of spermatogenesis: Characterization using a monoclonal antibody☆
[摘要] Inmammals,somedifferentiationantigensofspermatogenesisareautoantigens.Oneoftheseautoantigenshasnowbeenidentifiedandcharacterizedusingamonoclonalantibody,1B3.Theantibody-producingcellswerepreparedusingthespleencellsofamousewhichhadbeenimmunizedsyngeneicallywithvasdeferensspermatozoa.Evidenceispresentedthatthe1B3antigenresidesonasurfaceproteinorglycoproteinofmolecularweight28Kdaltons.Inabsorptionassaysusingavarietyofdifferentmousetissues,the1B3antigenwasdetectedonlyontesticularcellsandspermatozoa.Theantigenappearedonthecellsurfaceataspecificstageinspermatogenesis:itwasnotfoundoncellsoftheprepuberal,8-daymousetestis,butwasfirstseenonpachyteneprimaryspermatocytesarisingat14days.1B3antigenwasthenretainedonallpostpachytenecellswithinthetestis.Byimmunofluorescence,1B3antigenwasseentobepresentovertheentirecellsurfaceofspermatozoawithinthetestisandcaputepididymis,butcaudaepididymalandvasdeferensspermatozoashowedfluorescentlabellingonlyupontheanteriortipofthehead.Althoughtissuespecific,1B3antigenisnotspeciesspecific.Itwasfoundontesticularcellsofrat,rabbit,vole,andguineapig,butnotoftherhesusmonkey.Somepossibleapplicationsofmonoclonalantibodiestothestudyofspermatogenesisarediscussed.
