An interspersed region of the sea urchin genome represented in both maternal poly(A) RNA and embryo nuclear RNA☆

[摘要] Thesequenceorganizationandtranscriptionalexpressionofa17-kbregionoftheStrongylocentrotuspurpuratusgenomearedescribedinthisreport.Thisregionincludesatranscriptionunit,designated“gene88,”thatisrepresentedbyatypicallowprevalencematernalpoly(A)RNA.Thepredominantmaternalgene88transcriptisabout9.5kbinlength,andanalysisofthesequenceorganizationoftheclonedgenomicregionencodingthistranscriptshowsthatitcontainsahighdensityoflowandmoderatelyprevalentrepeatsequences,aswellas≥2kbofsingle-copysequence.GenomeblotscarriedoutwithDNAfromdifferentindividualseaurchinsshowthatseveralallelicformsofthissingle-copyregionexist.Thetranscribedsingle-copysequenceisonlyslightlylesspolymorphicthanistheaveragesingle-copysequenceoftheS.purpuratusgenome.Gene88single-copysequencesappeartoberepresentedinatleastthreesmallertranscriptsinthepolysomalRNAof16-cellembryos.ThenuclearRNAoflaterembryosalsocontainsgene88transcripts.RNAgelblotsindicatethatpolyadenylatedgene88transcriptsarealso9.5kbinlengthinblastulaandgastrulanuclearRNAsandthesetranscriptsareprobablycolinearwiththemajormaternalRNAtranscript.
