Induction of cell attachment and morphological differentiation in a pheochromocytoma cell line and embryonal sensory cells by the extracellular matrix☆
[摘要] Embryonicchickensensorycellsfromdorsalrootgangliaandaclonallineofpheochromocytomacells(PC-12)extendedneuronal-likeprocesseswithin24hrofseedingonanaturallyproduced,basementmembrane-likeextracellularmatrix(ECM)intheabsenceofnervegrowthfactor(NGF).PlatingonECMalsoinducedarapidcellattachmentandflatteningofthesecellsandsupportedthesurvivalofembryonicsensorycellsinprimarycultures.UnliketheeffectofNGFonPC-12cells,theECM-inducedmorphologicaldifferentiationwastransientandledtodisintegrationanddegenerationofprocessesbearingPC-12cells.TheECM-inducedmorphologicaldifferentiationwasnotinhibitedbyanti-NGFantibodies,andthecellsretainedtheirabilitytobindandinternalizeNGFinamannersimilartothatobservedonplastic.PC-12cellattachmentandflatteningoccurredondishescoatedwithcollagentypeIVinawaysimilartothatobservedonECM,butprecoatingthedisheswithfibronectinhadnoeffect.Extensionofcellprocesseswasnotinducedbyeithersubstrate.Morphologicaldifferentiationbutnottheinductionofcelladhesionandflatteningwasinhibitedbyeitherprefixationwithglutaraldehyde,oxidationwithperiodate,orpreexposuretoconcanavalinAoftheECM,suggestingthattheECMandinparticularitssugarmoietiesplayanactiveroleintheinductionofneuriteoutgrowth.ItissuggestedthatclosecontactwiththeECMprovideschemicalormechanicalcuesthatpermitcontactmediatedelongationanddirectedgrowthofbothembryonicandregeneratingnervefibers.
