Archenteron formation induced by ascorbate and α-ketoglutarate in sea urchin embryos kept in SO2−4-free artificial seawater☆
[摘要] Inpermanentblastulaeoftheseaurchin,whichwereobtainedbycultureinSO2−4-freeartificialseawaterfromthetimeoffertilization,ascorbateandα-ketoglutarate,activatorsofprotocollagenprolinehydroxylase,inducedtheformationofarchenteron.Byaddingeitherascorbateorα-ketoglutaratetotheSO2−4-freecultureat12hroffertilization,sphericalembryoswitharchenteronwereobtainedbysuccessive12hrculturesat20°C.Theembryosthusobtaineddidnotdeveloptoplutei.ArchenteronformationinducedbythesecompoundsinSO2−4-free-culturedembryos,aswellasinthenormalembryos,wasinhibitedbyα,α′-dipyridyl,aninhibitorofprotocollagenprolinehydroxylase.Glutamate,malate,citrate,andfumaratedidnotstimulatearchenteronformationinSO2−4-freeculturedembryos.IntheSO2−4-free-culturedembryosexposedto[14C]proline,considerableradioactivitywasfoundinhottrichloroaceticacid-extractableproteinsbuttheradioactivityof[14C]hydroxyprolineresidue,producedbyhydroxylationofprolineresidueofprotocollagen,wasmarkedlylowerthanthatinnormalembryos.Inthepresenceofascorbateandα-ketoglutarate,theradioactivityof[14C]hydroxyprolineresiduebecamehighandwasloweredbyα,α′-dipyridyl.Archenteronformationinducedbyascorbateandα-ketoglutarateintheembryoskeptinSO2−4-freeartificialseawaterprobablyresultsfromthestimulatedprotocollagenhydroxylation.
