Clonal analysis of vertebrate myogenesis: VII. Heritability of muscle colony type through sequential subclonal passages in vitro☆
[摘要] Musclecolony-forming(MCF)cellsfromvertebratelimbbudsshowstage-specificdifferencesinmediarequirementsandclonalmorphology(N.K.White,P.H.Bonner,D.R.Nelson,andS.D.Hauschka,1975,Develop.Biol.44,346–361).Thecelllineagerelationship,ifany,betweenthedifferentmyogeniccelltypesisunknown.ToinvestigatewhetherearlyMCFcellsconvertormatureintothelatetype,MCFcellsfromstage23–33chickembryolegbudsweregrowninvitroandsequentiallysubcloned.Resultsindicatethatcolonytype,nomatterwhatthestageoforigin,isheritableinvitrothroughatleastfivepassages(45invitrodoublings)oruntilthesubclonesreachproliferativesenescence.TheseresultsarediscussedwithrespecttovariousalternativesforthegenerationofMCFcelldiversityduringlimbdevelopment.
