Transdifferentiation and regeneration in vitro☆
[摘要] StriatedmuscletissueandendodermcanbeisolatedfromtheanthomedusaPodocorynecarnea.Theisolatesareuncontaminatedbyothercelltypesandcanbecultivatedinartificialseawaterformonthswithoutundergoingautonomousregeneration.However,iftheendodermiscombinedwithcollagenase-treatedstriatedmuscle,aregenerationprocessisinitiatedwhichleadstotheformationofthesexualandfeedingorgan(manubrium)ofthemedusa.Theoriginalendodermandstriatedmusclearereplacedintheregeneratebyatleastsevennewcelltypes,includinggametes.Labelingexperimentswith[3H]thymidineandexperimentsinwhichmitosisisinhibitedineitherthestriatedmuscleortheendodermwithmitomycinCdemonstratethatthestriatedmuscleisabletotransdifferentiateintoallthecelltypesfoundintheregenerate.Withthepossibleexceptionofectodermalsmoothmusclethisstatementisalsovalidfortheendoderm.
