Cell fusion competence and its induction in Physarum polycephalum and Didymium iridis☆
[摘要] InheterothallicMyxomycetes,diploidplasmodiaarisewhenhaploidamoebaeoftwodifferentmatingtypesareculturedtogether.Inthismatingprocess,theamoebaefuseinpairs,andtheresultingzygotesdevelopdirectlyintoplasmodia.Ithasbeenshownpreviouslythatplasmodiastarttoforminthisfashiononlywhenthegrowingamoebaeinamixedculturereachacriticaldensity.Wehaveinvestigatedthecellularbasisofthisphenomenonbygrowingamoebaeofdifferentmatingtypeseparatelyfromoneanotherandthenmixingthemtotesttheirmatingability.Amoebaefromculturesaboveandbelowthecriticaldensitywere,respectively,competentandincompetenttomate.Furthermore,bothpartnershadtobecompetentinorderformatingtooccur.Nobinucleatecellswereformedinmixturesofincompetentamoebae,indicatingthattheyfailedtofusewithoneanother.Incompetentamoebaegrowingatlowdensityonfilterswith0.2-μmporesbecamecompetentwhenthefilterswereplacedondenseculturesofamoebae.Wesuggestthatamoebaereleaseafilter-transmissiblematerialthataccumulatesduringgrowthandinducesthecellstobecomefusioncompetent.
