Cell surface polarization, tight junctions, and eccentric inner cells characterize human teratocarcinoma embryoid bodies☆
[摘要] ThesestudiesdemonstratethathumanembryoidbodiescandevelopinvitrofromhumanteratocarcinomacelllineHT-H.Furthermore,scanningandtransmissionelectronmicroscopyrevealthattheseembryoidbodiesresemblethepreimplantationembryomorethanmouseembryoidbodieswhicharemultilayered.Thisresemblanceisbasedon(1)aneccentricallylocatedgroupofinnercellswithinathin-walledcyst,(2)apicaljunctioncomplexeswithtight,gap,anddesmosomaljunctions,aswellasintermediatefilamentsassociatedwiththeoutercelllayerandnottheinnercells,and(3)mitochondriawithanextremelydensematrixandfewplate-likecristae.Observationsofthecellsurfaceindicatethatmicrovillihaveapolarizeddistributionandarelocalizedtotwoareas(centrallyandatintercellularborders)ontheapicalcellsurfaceofcompactedaggregates.Immunosurgeryofcysticembryoidbodiesindicatesthatatleasthalfofthemarecharacterizedbyinnercellsenclosedbyanimpermeableoutercelllayer.ThesedevelopmentalsimilaritieswiththepreimplantationmouseembryosuggestthatatleastsomeHT-Hteratocarcinomacellsexpressanembryo-likeprogramformulticellularorganization.HT-Hembryoidbodyorganizationischaracteristicofthepreimplantationembryoincontrasttotheorganizationreportedformouseembryoidbodieswhichresemblesthepostimplantationinnercellmass.
