Cleavage stage mouse embryos share a common cell adhesion system with teratocarcinoma cells☆
[摘要] Weexaminedsimilaritiesinadhesivepropertiesofmousecleavingembryosatone-toeight-cellstagesandofteratocarcinomacellsbyaggregationstudies.TeratocarcinomacellsandfibroblasticcellshaveaCa2+-dependentcell-celladhesionsite(CDS),whichisresistanttotrypsininthepresenceofCa2+butsensitiveintheabsenceofCa2+.WhenseveralembryostreatedwithtrypsininthepresenceofCa2+(TC)werekeptincontactwitheachother,theyfusedintoasingleaggregateinthemediumwithCa2+butnotwithoutCa2+.EmbryostreatedwithtrypsinintheabsenceofCa2+(TE)didnotshowsuchCa2+-dependentaggregation.AggregationofTC-treatedembryoswasinhibitedbyFabfragmentsofantibodyraisedagainstTC-treatedteratocarcinomaF9cells.Theaggregation-inhibitoryeffectoftheFabwasremovedbyabsorptionwithTC-treatedteratocarcinomacells,butnotwithTE-treatedteratocarcinomacells.Thiseffectwasnotremovedbyabsorptionwithfibroblastsandsomeothertissuecells.TC-treatedembryosadheredtoTC-treatedteratocarcinomacells,butnottoTC-treatedfibroblasticcells.TheseresultssuggestthatearlymouseembryosshareacommonCDSmoleculewithteratocarcinomacellsbutnotwithfibroblasticcells.
