Accumulation of individual histone mRNAs during embryogenesis of the sea urchin Strongylocentrotus purpuratus☆
[摘要] WehavedeterminedthatthestockpileofmaternalhistonemRNAsforbothH2BandH3ineggsoftheseaurchinStrongylocentrotuspurpuratuscorrespondsto0.8–1millionmoleculesperegg.Thisvalueisasomewhatsmallerquantitythanhadpreviouslybeenestimated.Ontheotherhand,theincreasebroughtaboutbyembryonicmRNAsynthesisis10-to12-fold,ratherthan4-fold.FurtheranalysisofthetimecourseofaccumulationofthesemRNAssuggeststhathistonegeneexpressioninearlydevelopmentinvolvesacomplexinteractionbetweenactivationofstockpiledmaternalmessengers,fluctuatingratesofhistonegenetranscription,andalterationsofhistonemRNAstability.ThematernalpoolofH1mRNAisconsiderablysmaller(80,000molecules).Duringcleavage,itpartiallycatchesupandpeaksatabouthalfthevalueforH2BandH3.WealsoexaminedprevitellogenicoocytesforthepresenceofhistonemRNAsbythesamemethodsandwereunabletodetectany.ThusthestockpileofmaternalhistonemRNAsmustaccumulateaftertheprevitellogenicstageofoogenesis.
