Development of cholinergic neurons of the rat retina
[摘要] Biochemicalandelectrophysiologicalfeaturesofcholinergicneuronsfromthedevelopingratretinawereanalyzedinanattempttoidentifyimportantperiodsinthematurationoftheseneurons.Acetylcholinesynthesiswasassayedinintact,isolatedretinas;cholineacetyltransferaseactivitywasmeasuredinhomogenates.Spontaneousandevokedacetylcholinereleaseandcertainaspectsofsynapseformationwereexaminedinaretina-musclecellculturesystem.Inthissystem,retinalcellsfromratsofdifferentagesweredissociatedwithtrypsinandaddedtopreviouslyculturedratstriatedmusclecellswhichservedaspostsynaptictargetsforcholinergicneurons.Theresultsindicatethattwodevelopmentalperiodscanbedescribed.Thefirststageoccursduringthefinalweekofgestationwellbeforemorphologicalsignsofsynapsesappear.Duringthisearlyperiod,cholinergicneuronsacquiretheabilitytosynthesizeandreleaseacetylcholineandtoformfunctionalsynapses.Thesedevelopingneuronsprogressivelylosetheirabilitytoformandtomaintainsynapseswithaninappropriatetarget.Asecondstageofmaturationbeginsattheendofthefirstpostnatalweek.Inthisphase,dramaticincreasesinacetylcholinesynthesis,cholineacetyltransferaseactivity,andhigh-affinitycholineuptakearecoupledtemporallywiththemorphologicaldifferentiationofsynapses.
