Chick ciliary ganglion in dissociated cell culture: I. Cholinergic properties☆
[摘要] Culturedciliaryganglionneuronshavebeenusedtostudycholinergicmetabolismandtocharacterizeaneurotrophicfactorforparasympatheticcells.Optimalconditionswereestablishedfor(i)cholineacetyltransferase(CAT)activityand[3H]acetylcholine(ACh)synthesisfrom[3H]choline,and(ii)forthecultureoftheseneuronsinthevirtualabsenceofnonneuronalcells.Eyeextract,humanserum,andconditionedmediafromgliomaandstriatedmusclecellswerefoundtobeequallyactiveasneurotrophicfactorsforneuronsculturedalone;thecriteriaweresurvival,CATlevelsand[3H]AChsynthesisforneuronscultured8days.Whencoculturedwithfibroblasts,ganglionicnonneuronalcellsormyotubes,neuronsincreasedonlyslightlytheirsynthesisof[3H]AChascomparedtoneuronsalone;therefore,thepresenceofothercelltypesdidnotinfluencethecholinergicproperties.Inallcases,elevatedKClconcentrationswereessentialformaximumsurvivalanddevelopmentofthecholinergicpropertiesoftheneurons.
