Accumulation of the early histone messenger RNAs during the development of Strongylocentrotus purpuratus☆
[摘要] ThedevelopmentalprofileofamountsofthemRNAsfortheearlyhistoneswasdetermined.Seaurchinembryos(Strongylocentrotuspurpuratus)werelabeledwith3Hnucleoside,andtheRNAwasextractedandfractionatedbyelectrophoresisonpolyacrylamidegels.RadioactivityintheresolvedmRNAbandswasdeterminedandconvertedtomolarquantitiesbyknowledgeoftheprecursorpool-specificactivity.Betweenthe16-andthe200-cellstages7–10×106moleculesofeachcorehistonemRNA,and2.5×105moleculesofH1mRNAaccumulate.DuringthesubsequentfewhoursofcleavagetheaccumulatedmRNAsdisappearwithahalf-lifeof1.5–2hr.ItisarguedthatmRNAhalf-lifemayberegulatedduringcleavage.
