Nerve growth factor has both mitogenic and antimitogenic activity☆
[摘要] Thepresentstudydemonstratesthatnervegrowthfactor(NGF)possessesbothantimitogenicandmitogenicactivities.Tothisend,wehaveemployedclonalPC12ratpheochromocytomacellsandtwoPC12variantsublines,U2andU7.WhenPC12cellsareexposedtoNGFinculturemediathatareotherwiseeitherpermissive(15%serum)orrestrictive(1%serum)forproliferation,neuronaldifferentiationoccursandmitosisceases.VariantlinesofPC12cellshavebeenselectedthatcontinuetoproliferateinthepresenceofNGFinpermissivemediumbutwhichneverthelessretainNGFreceptorsandcertainNGFresponses.IncontrasttotheparentPC12cells,whensuchvariantswereexposedtoNGFingrowth-restrictivemedia,cellproliferationwasmarkedlystimulated.ThemitogenicactivityofNGFwasdetectableat0.1ng/ml(4pM)andwasmaximalat3ng/ml(100pM).PossiblecontaminationoftheNGFpreparationbyepidermalgrowthfactor(EGF)ormitogenicproteolyticenzymeswasruledoutbytheuseofanti-EGFanddiisopropylfluoro-phosphate,respectively.ThesefindingsshowthatNGFsharesthecapacitytostimulatecelldivisionwithavarietyofotherpeptidehormonesandsuggestthatthemitogenicactivityofNGFcouldplayaroleindevelopmentoftheperipheralnervoussystemaswellasinpromotionofinvivogrowthofcertainneuralcrest-derivedneoplasms.
