situhybridizationandNorthernblotanalysisindicatesthattroponinIcisfirstexpressedintailbudembryos(stage28)aboutthetimeofthefirstcytologicalheartdifferentiationandabout24hrbeforebeatingtissueisobserved.Wehaveusedthismarkertoexamineabnormalheartmorphogenesisinembryostreatedwithretinoicacidandlithium.Whenretinoicacidisadministeredtoembryospriortoheartspecification,hearttissueisreducedandoftencompletelyablated.Whenembryosaretreatedafterheartspecification,butbeforetheheartprimordiummigratestotheventralmidline,themigrationisunaffectedbutsmaller,abnormalheartsresult.Lithiumtreatmentofcleavagestageembryoscausesanincreaseinhearttissue.Inseverelydorsalizedembryos,hearttissuecanbefoundaroundtheentireembryowiththeexceptionofasmallgapatthemostdorsalpoint.Thisgapindicatesthatmigrationofthehearttotheventralmidlinedoesnotoccurintheseembryos.Laterindevelopment,acentrallylocated,beatingheartisobservedindorsalizedembryos.Thetimingofitsappearancesuggeststhatitisformedbymovementsnormallyassociatedwithheartmorphogenesisratherthanmigration.
