A possible cause of termination of cell growth in the cellular slime mold Dictyostelium discoideum☆
[摘要] Amacromolecularinhibitorofcelldivisioncanbewashedoffexponentiallygrowingaswellasstationaryslimemoldamoebae.However,stationarycellsappeartohavefourtimesasmuchmitoticinhibitingactivityasthesamenumberofexponentialcells.Removalofsuchaninhibitorbywashingcellsduringexponentialgrowthdelayedtheonsetofstationaryphaseandresultedincultureswithafinalcelldensitydoublethatnormallyattained.Additionofapartiallypurifiedfractionofdivisioninhibitortorapidlygrowingcellsresultsintheabruptterminationofcelldivision.Itisalsodemonstratedthatconditionedmediumcontainssufficientnutrientsforcellgrowth.Therefore,thedivisioninhibitormayplayasignificantroleinthecessationofgrowth.Proteincontentofamoebaeincreasesduringvegetativegrowth,reachingamaximumatstationaryphase.TheeffectofcyclicAMPoncelldivisionwasexamined.Ahypothesisonthemechanismofactionandadaptivesignificanceofthedivisioninhibitorisdiscussed:Amacromolecularmitoticinhibitorbeginstoaccumulateonthecellsurfaceduringvegetativegrowth.Whentheinhibitorsaturatesthecellsurface,mitosisisprevented.Thisinhibitionmayoccurthroughactivationofadenylcyclase.
