Inhibitory effect on limb morphogenesis by cells of the polarizing zone coaggregated with pre- or postaxial wing bud mesoderm☆
[摘要] Theinfluenceofcellsofthepolarizingzonemesodermonthemorphogenesisofrecombinantchicklimbswasstudied.Therecombinantbudswerecomposedoflegbudectodermanddifferentregionsofthewingbudmesoderm,whichhadbeendissociatedandreaggregated.Inanycasewherethepolarizingzonemesodermwascoaggregatedwiththewingmesodermthemorphogeneticcapabilitiesoftherecombinantwerereduced.Thiswasthecasewithpostaxialmesoderm,preaxialmesodermpluspolarizingtissue,andpostaxialmesodermfromwhichapieceofthenonpolarizingmesoderm(comparableinsizetothepolarizingzone)hadbeenremoved.Allofthesegaveoutgrowthswithdigitsinonlyaverylowpercentageofcases.Incontrast,thoserecombinantswithoutpolarizingmesodermdevelopedoutgrowthswithdigitsinahighpercentageofcases,indicatinggoodmorphogenesis.Finally,ifthepolarizingzonewereremovedpriortodissociation,therecombinantlimb,composedofthetotalremainingwingbudmesodermpluslegbudectoderm,exhibitedahigherpercentageofcompletemorphogenesisthanifthepolarizingzonehadbeenpartoftherecombinant.Itisclearthatcellsofthepolarizingzone,whendissociated,andcoaggregatedwithwingmesoderm,areinhibitorytothemorphogeneticperformanceofthatmesodermintherecombinantlimbsituation.
