Ultrastructural change of melanosomes associated with agouti pattern formation in mouse hair☆
[摘要] Wehavestudiedthestructuralalterationofmelanosomesinthemelanocytesofagoutimicewhosegeneticcharacteristicistoproduceeumelaninandphaeomelaninalternatelyinasinglehairbulb.Melanocytesofhairbulbsfrom1to2dayoldmiceoftheblackphasewereobservedtocontainrod-shapedmelanosomesoftheeumelanintype(eumelanosome).Inthemelanocytesofthehairbulbsfrom4to6-dayoldskin,whichexclusivelycontainphaeomelanin,sphericalmelanosomes(phaeomelanosomes)wereseen.Ontheotherhand,themiceofthetransitionalphasefromblacktoyellowpossessedmelanocytesthatcontainedbotheumelanosomesandphaeomelanosomeswithinasinglecell.Thisresultindicatesthattheshiftfromtheeumelaninformationtothephaeomelaninformationorviceversainagoutihairoccurswithinasinglemelanocyte.Weobservedmultivesicularbodiesinboththeagoutimelanocytesoftheyellowphaseandthegenotypicallyyellowmelanocytes.Thesebodiesareconsideredtobetheprecursorofthephaeomelanin-containingmelanosome.TheyaresometimesobservedtohavecontinuitywithE.R.suggestingthatthemelanosomesarederivedfromE.R.inthephaeomelanin-formingmelanocytes.
