Alterations in DNA-dependent RNA polymerase activity during the development of the tobacco hornworm, Manduca sexta☆
[摘要] RNApolymerasesI,IIandIIIhavebeendetectedintheextractsoffatbodyandintegumentofthetobaccohornworm,andtheiractivityduringlarval-pupal-adultmetamorphosishasbeenmeasured.TotalRNApolymeraseactivityofbothtissuesreachesapeakjustpriortothewanderingstageofthefifthinstarlarva.Theenzymeactivityoftheintegumentdeclinesthereafterwhile,inthefatbody,achangeinthecellularcompartmentalizationofenzymeactivityoccursduringdevelopment.ThisisindicatedbytheobservationsthatRNApolymeraseactivity,whichwaspredominantlyinthesolublefractionupuntiltheonsetofthewanderingstage,declinesrapidlyduringthewanderingstagewhileRNApolymeraseactivityintheinsoluble-pelletfractionincreases.Asteady-statelevelisreachedjustpriortopupation,andtheenzymeactivityremainsatthatlevelduringpharateadultdevelopment.Theα-amanitin-sensitiveenzymeappearstoberesponsibleformostoftheRNApolymeraseactivityduringlarvallife.ThefindingsthatthepeakofRNApolymeraseactivityinbothtissuesandthesubsequentchangesincompartmentalizationinthefatbodyoccurcoincidentallywiththefirstsurgeofα-ecdysonereleasebytheprothoracicglandsraisesthepossibilitythatcontrolofRNApolymeraseactivitymaybehumorallymediated.
