Appearance and distribution of carbohydrate-rich macromolecules on the epithelial surface of the developing rat palatal shelf
[摘要] Thesynthesisandappearanceofcarbohydrate-richmacromoleculesbyepithelialcellsofthedevelopingsecondarypalatewasexaminedwithconcanavalinA(CONA)bindingand[3H]glucosaminelabeling.Theamountof[125I]CONAboundtotheepithelialsurfaceoftheratpalatalshelfinvitroincreasedfromday15ofgestationtoday16wheninitialadhesiontotheoppositeshelfoccursinvivo.VisualizationofCONAbindingbyelectronmicroscopyusingtheperoxidasemethodrevealedadramaticincreaseinbindingbetweendays15and16ofgestation,mostapparentonthemedial-edgeepithelialsurface.Theincorporationinvivoof[3H]glucosamineduringthisperiodintothemedial-edgeepithelialcellswasdetectedwithautoradiography.Theseresultsshowthataglycoprotein-richsurfacematerialappearsonthesuperficialcellsofthemedial-edgeepitheliumpriortoadhesionoftheapposingshelves.
