The amoebal cell of Physarum polycephalum: Colony formation and growth☆
[摘要] TwostagesofcolonygrowthwereobservedduringmicroscopicstudiesofPhysarumpolycephalumamoebae.Duringthefirststage,“spreadinggrowth,â€thecolonyiscomposedofdispersedsinglecells.Duringthesecondstage,“aggregategrowth,â€mostoftheactivecellsinacolonyareaggregatedinaringatthecolonyboundary.Measurementsofcellmovementasafunctionofbacterialconcentrationindicatethat,duringbothspreadingandaggregategrowth,cellmovementsarenotaffectedbychangesinbacterialconcentrationbutthatthetransitionfromspreadingtoaggregategrowthoccursearlieronplateswithlowerbacterialconcentrations.Theseresultsindicatethatautonomouscharacteristicsoftheamoebaearemoreimportantforthedeterminationofcolonyformthanlocalvariationsintheconcentrationsofnutrients.Thegeneticdeterminationofcolonyformisdemonstratedbytheexistenceofmutantsthatdisplayspecificalterationsincolonymorphology.Becausetheaggregateringsofthesemutantsmoveatanincreasedrate,mutantclonesappearasvariantsectorsofwild-typecolonies.Theincreasedrateofmutantringmovementsuggeststhatthisselectionmethodmaybeausefultechniqueforisolatingmutantmyxamoebaewithdefectsinmovementandbehavior.
