The isolation of intact cortical granules from sea urchin eggs: Calcium ions trigger granule discharge☆
[摘要] Asimpleprocedureisdescribedforisolatingpreparationsofcorticalgranulelayersfromseaurchineggs.Theisolatedgranulesarestructurallyintactasrevealedbyscanningandtransmissionelectronmicroscopy.WhenCa2+ispresent,theisolatedgranulesinstantaneouslydischargetheircontents.ThesiteofactionofCa2+mayresideinthemembraneofthegranule.Procaine,acompetitiveinhibitorofCa2+bindingtomembranes,completelyblocksthedischargeofcorticalgranulesthatnormallyoccursatfertilization.Theresultssupportthehypothesisthatonceinitiated,thepropagationofcorticalgranuledischargespreadsasanautocatalyticwaveinwhichdischarginggranulesreleaseCa2+throughtheirmembraneswhichinturntriggersthedischargeofadjacentgranules.
