Effects of differences in mitotic activity, stage of cell cycle, and degree of specialization of donor cells on nuclear transplantation in Xenopus laevis
[摘要] TheabilityofnucleioftwokindsofcellsfromtheintestinalepitheliumofadultX.laevistosupportdevelopmentofrecipientoocyteswascompared.Cellssituatedbetweenthelongitudinalfoldsofthegutwall(troughcells)arefunctionallyandstructurallyunspecialized,divideasynchronously,andindividualcellsarethereforeindifferentphasesofthecellcycle.Cellsfromthetopsofthefolds(crestcells)aremitoticallyinactive,allinthesamephaseofthecellcycle,andarefunctionallyandstructurallyspecialized.Despitethesedifferences,thepatternsofdevelopmentofrecipientoocytestowhichthesenucleiweretransplantedwereidentical.Thedevelopmentofprimaryandserialtransplantsofgutnucleiwascomparedwiththatofcontrolblastulanucleiandfertilizedeggswhichhadbeenirradiatedandprickedtosimulatetransplantation.Thecomparisonshowedthatthemajorfactorrestrictingdevelopmentoftransplantswaswhetherthecellshadbeguntodifferentiate.Theserestrictionswerestablethroughserialtransplantation.Theireffectwastodecreasetheproportionofrecipientoocyteswhichinitiatedcleavageandwhichcontinuedtocleavenormallyandtoincreasethefrequencyofabnormalitiesaftergastrula.
