Clonal analysis of vertebrate myogenesis: IV. Medium-dependent classification of colony-forming cells☆
[摘要] Thecelllineageofchicklegmusclebetween3and12daysofdevelopmenthasbeenstudiedbyuseofaninvitroclonalassay.Theassaypermitsdistinctionstobemadeamongvarioustypesofmuscle-colony-formingcells(MCFcells)onthebasisoftheirmediumrequirementsandclonalmorphology.ResultssuggestthesequentialoccurrenceofatleastfourtypesofMCFcells,threeofwhichrequireconditionedmediumfortheirdifferentiationandoneofwhichcanformdifferentiatedcoloniesinfreshmedium.Thenatureofthe“conditionedmediumeffect”wasfurtherinvestigatedbytheuseofmedium-switchexperiments.Bythisprocessitwasshownthatthesamepopulationsofcolony-formingcellsattachandgrowinfreshandconditionedmediumandthatthedifferentiationofcoloniesderivedfromconditioned-medium-requiringmyoblastsispermittedbybriefexposuretoconditionedmediumfollowedbycultureinfreshmedium.Furtherinvestigationindicatedthatduringbriefexposuretoconditionedmediumthegelatin-coatedpetriplatesurfaceisalteredsuchthatdifferentiationofconditioned-medium-requiringcoloniesisallowed.Weconcludethattheconditionedmediumeffectinvolvesasurface-mediatedinteractionbetweenmyoblastsandoneormoreconditionedmediumcomponents.
