Corneal development: V. Treatment of five-day-old embryos of domestic fowl with 6-diazo-5-oxo-l-norleucine (DON)☆
[摘要] TheembryogenesisofthecornealstromaofthedomesticfowlwasstudiedfollowingtheinjectionoftheglutamineanalogDON(6-diazo-5-oxo-l-norleucine)intothechorioallantoicveinsof5-day-oldchickembryos.The44survivorswerecilledbetween1hrand13daysfollowinginjection.Theircorneaswereexaminedhistologicallyandcomparedwiththoseofuntreatedanimalsofsimilarage.Alloftheexperimentally-treatedcorneasexamineddevelopedabnormally.Thedefectsincluded:atemporarydisappearanceofportionsoftheendothelium;thedepositionofdisorderedarraysofcollagenfibersbeneaththecornealepithelium,includingareversalinthedirectionofrotationoftheaxesofaffectedportionsofthestromallamellae;theappearanceofstromalcysts;andtheaccumulation,beginning6daysafterinjection,ofpoolsofGomori-silver-positivematerialwithintheepithelium.AbnormalitiesincornealdevelopmentfollowingtreatmentwithDONwerecomparedwiththosepreviouslyobtainedfollowingadministrationofl-azetidine-2-carboxylicacid.Thefindingsdemonstratedthat:(1)thecharacteristic,progressiverotationoffibrilorientationwhichnormallyoccursintheouterlamellaeoftheavian,corneal,primarystromaisnotarigidly-determinedconfigurationsinceitsdirectioncanbereversedconsistentlyfollowingtreatmentwithDON;and(2)theprimarystromadictatesthecollagenousstructureofthesecondarystromadepositedwithinit.
