Biochemical studies of growing mouse oocytes: Preparation of oocytes and analysis of glucose-6-phosphate dehydrogenase and lactate dehydrogenase activities

[摘要] Oocytesatseveralstagesofgrowthwereisolatedbyenzymaticdigestionofovariesfrominfantmice.Theseoocyteswerefreeoffolliclecellsandwereobtainedinsufficientnumberstopermitdirectbiochemicalanalysisofglucose-6-phosphatedehydrogenase(G6PD)andlactatedehydrogenase(LDH)activities.Bothenzymesincreaseintotalactivityastheoocytegrows.However,whilethespecificactivityofG6PDremainsconstantuptoanoocytediameterof80–85μm,thespecificactivityofLDHincreasesfour-fold.ThespecificactivityofG6PDandespeciallyLDHdeclinesinoocytesover80–85μmindiameter,suggestingthatthesynthesisoftheseenzymesisalmostcompletedatthisstageofgrowth.
