Thyroxine and palatal development in rat embryos
[摘要] Theplacentaltransportandpalatallocalizationofl-thyroxine-125IwasstudiedinSprague-Dawleyratembryosages13and14daysinvivoand14–16daysinvitro.Amnioticfluid,placentaand(bylateday14)embryonic/palatalandliverareaswereassayedbyscintillationcountingandproteinanalysis.Radioactivitywasfoundinamnioticfluidasearlyas13daysinvivo.Asmallbutconsistentamountofradioactivityabovecontrollevelswasfoundintheembryonicpalatalandliverareas.Autoradiographsofthin-layerchromatographsshowedthatmostoftheradioactivelabelwasatthethyroxineareainboth13-and14-dayinvivoand15-dayinvitroamnioticfluidpools.Asmallamountofradioactivitywaspresentinthediiodothyronineareainboth.Someactivitywasalsopresentinthetriiodothyronineareainthe13-and14-daysamples.Nolabelledinorganiciodidewasdetectable.Thethyroidglandinratdoesnotbegintofunctionuntil17daysinutero.Accordingly,thelabelledthyroxinewasexogenous.Thepresenceoflabellingintheembryonicpalatesuggestsapossibleinvolvementofthishormoneinpalatalembryogenesis.
