Differentiation and maturation of cultured fetal rat jejunum☆
[摘要] Todeterminewhetherdifferentiationandmaturationofmammalianintestinalmucosarequireinfluencesavailableonlyinvivoorwhethertheycanoccurinvitro,fetalratjejunumwasculturedinchemicallydefinedmediumusingorganculturemethodology.Segmentsofjejunumfrom18-dayfetalratswereculturedinmodifiedLiebowitzL-15mediuminroomairat37°C.Segmentsharvestedafter24,48,and72hrofculturewereexaminedbylightandelectronmicroscopy.Unculturedjejunumfrom18-dayfetuseshadeithernoorveryfewrudimentaryvilliandwaslinedlargelybyundifferentiatedstratifiedepithelium.Gobletcellswerenotseen.Incontrast,villiwerepresentinthemajorityof24-hrcultures,andsimplecolumnarratherthanstratifiedepitheliumpredominated.After48and72hr,villiwerepresentinover90%ofculturedjejunalsegmentsandstratifiedepitheliumhaddisappeared.Gobletcellswereseeninjejunalsegmentscultured48hrorlongerin47of74fetuses.Electronmicroscopyfurtherdocumentedprogressivedifferentiationoftheepitheliumduringculture.Microvilliincreasedinnumberandheight,aterminalwebdevelopedintheapicalcytoplasmandthenumberofapicalvesicles,mitochondriaandformedelementsofendoplasmicreticulumincreasedinabsorptivecells.Jejunallactaseandalkalinephosphataseactivityincreasednine-andsevenfold,respectively,during72hrofculture,whiletheactivityofthemitochondrialenzyme,ornithinecarbamoyltransferase,increasedfourfold.Theseobservationsindicatethatjejunumfrom18-dayfetalratscanbeculturedinvitroforatleast72hrinchemicallydefinedmediumandthat,duringculture,maturationofthejejunalmucosatakesplacewiththeappearanceofvilli,conversionofstratifiedtocolumnarepitheliumanddifferentiationofindividualepithelialcells.
