Changes in amino acid uptake and protein synthesis of bullfrog tadpole liver and tail tissues during triiodothyronine-induced metamorphosis☆
[摘要] Theeffectoftriiodothyronine(T3′)ontheuptakeofseveralaminoacidsintotheaminoacidpoolsandintoproteinsofRanacatesbeianatadpoleliverandtailmuscleandtailfinhasbeenstudied.Labelingofthealanineandglycinepoolwasstimulatedinthelivermorethantheleucinepool.AfterexposuretoT3for3days,uptakeofα-aminoisobutyricacid(atransportmodelsubstrate)intoliverwasstimulatedabout55%.IntailtissuesuptakeofleucinewasstimulatedbutuptakeofalaninewasdepressedbyT3.IncorporationofleucineandalanineintotissueproteinwasstimulatedintheliverbutinhibitedintailtissuesafterT3injection.ChangesinothermacromoleculesandATPandADPlevelsinliverandtailmusclewerealsoinvestigatedduringinducedmetamorphosis.Intheliver,thetotalDNAcontentdidnotchange,buttheRNAandproteincontentperliverincreasedsignificantly.TheincreaseinRNA/DNAandprotein/DNAratios,suggestedthatlivercellsunderwenthypertrophyduringinducedmetamorphosis.TheATPlevelshowedatransientdecreaseafter3daysofT3treatment.Intailmuscle,proteinandRNAcontentdecreasedasthemuscleregressed,buttheDNAcontentandATPlevelremainedunchangedthroughouttheexperimentalperiod.
