Long term organ culture of rat liver rudiment in a synthetic medium: Morphological and biochemical development☆
[摘要] Embryonicratliverrudimentsweregrownandmaintainedinachemicallydefinedmediumforupto16weeks.Liverexplantsgrewanddevelopedliverplates,bilecanaliculiandductules.Livercellsdifferentiatedand,bytheendofthesecondcultureweek,revealedcytologicalandultrastructuralfeaturesofadultratlivercells.Glycogensynthetaseactivityappearedandreachedaplateauwithin2weeks.Plasmaproteinsproducedbytheexplantsappearedsequentiallyintheculturemedium.Theirrelativeconcentrationsinthemediumincreasedprogressivelytoaplateauandtheypersistedthroughoutthecultureperiod.Albuminwaspresentintraceamountsduringearlydaysofculture.Itsrelativeconcentrationincreasedanditbecamethemajorproteinconstituentofthemediumbythesecondweek.Transferrin,α1M-globulin,haptoglobin,β1c-andα1B-globulinappearedinthatorder.Thethreefibrinogensubunitswereidentifiedduringthefirstweekofculture.Fetalproteinswereidentifiedduringtheearlycultureperiod,themostprominentbeingα-fetoprotein.Therelativeconcentrationoffetalproteinsdeclinedsteadilyasthecultureprogressed.
