The behavior of fetal canine cardiac cells in culture: Synthesis and phosphorylation of myosin☆
[摘要] Thisstudydescribesthefirstinvitroculturingofcaninecardiaccells.Caninecardiacmyosinwhichwassynthesizedina14-daytissueculture,basedonl-[3H]leucineincorporation,wasprecipitatedwithgoatγGantimyosin(cardiac-specific)andanalyzedondodecylsulfategels;thespecificactivityofthehighlypurifiedmyosinchainswasdetermined.Incorporationof32PO4wassimilarlyanalyzed.Thecomparativedegreeofsynthesisandphosphorylationofmyosinchains,occurringinculture,wasthesameasthatobtainedinvivo.Bothl-[3H]leucineand32PO4incorporationwereinhibitedbyadditionofcycloheximidetotheculturemedium.Removalof32PO4frommyosinheavychainswithbasetreatmentindicatedthepresenceofphosphoserineand/orphosphothreonineincaninecardiacmyosinheavychains.Myosinsfromfetalandadultcaninecardiactissuewereimmunologicallyidenticalwitheachotherandwiththeculturedfetaltissue;allhadsimilarmyosinATPaseactivityandthedegreeofheavychainphosphorylationwassimilar.Thetissueandtechniquesusedheregaveahighyieldofcardiacmyocytesbasedprincipallyonsynthesisofcardiac-specificmyosin.
