Contribution of a change in mRNA half-life to the accumulation of the tissue-specific S-100 protein during postnatal development of the mouse brain☆
[摘要] TheS-100proteinaccumulatesrapidlyinthemousebrainbetween15and21daysofpostnataldevelopment.Theaccumulationofthisproteinisbroughtaboutmainlybyanincreasedrateofitssynthesis.Thepresentstudyfocusesonattemptstodetermineifachangeinthehalf-lifeofmessengerRNA(mRNA)isinvolvedinbringingabouttheincreasedrateofsynthesisoftheS-100protein.UtilizingtheinhibitionofRNAsynthesisbyactinomycinD,wewereabletoshowthatthehalflifeofmRNAincreasesconcurrentlywithanincreaseintherateofsynthesisoftheS-100protein.UtilizationofactinomycinDdoespresenthazardsininterpretationofresultsonmRNAstability;experimentswereperformedtodetermineiftheresultsobtainedwereduetosideeffectsofthedrug.Asfarascouldbedetermined,thepossiblesideeffectsofactinomycinDdidnotaffectourresults.
