Heparan sulfate biosynthesis by embryonic tissues and primary fibroblast populations☆
[摘要] Fibroblastsfromcornea,heart,andskinofday14embryonicchicksdemonstratetheabilitytomakeheparansulfate-likepolysaccharidewhenexaminedduringthe10hrperiodimmediatelyfollowingtheirremovalfromtheembryo.Boththewholetissuesfromwhichthesefibroblastsareisolatedandthefibroblastsgrownfor2–5weeksinvitroalsosynthesizeheparansulfate.Duringtheirfirstfewdaysinvitro,thethreefibroblastpopulationsdisplayincreasingratesof[35S]-sulfateandd-[1-3H]-Glucosamineincorporationintoglycosaminoglycansandsharpfluctuationsofthoserates,yetthepercentageoftotal[35S]-sulfateincorporatedintoheparansulfate-likepolysaccharideandthedistributionofthispolysaccharidebetweencellsandnutrientmediumdonotchangesignificantly.Duringtheirfirst48hrinvitro,skinfibroblasts,butnotthosefromcorneaorheart,showsteadilydecreasingdiscrepanciesbetweentheproportionsof[35S]-sulfateandd-[1-3H]-Glucosamineincorporatedintoheparansulfate,suggestingasharpdeclineinthesynthesisofnonsulfatedglycosaminoglycans.ThesedatasupportthehypothesisofKraemerthanmanycell-typesinvivomaynormallymakeheparansulfate.Thedatalargelyeliminatethehypothesisthatthebiosynthesisofthispolysaccharideisselectivelystimulatedasembryoniccellsadapttogrowthinvitro.
